The peptide contains 4 tandem repeats of the tetrapeptide QQPP. The sequence is from a C-terminal region of the circumsporozoite protein containing 8 approximate tandem repeats of the 4-mer QQPP.
Mice of strains BALB/c, B6.D2, and B10.D2 immunized with the epitope were not significantly protected from infection after challenge with 500 live sporozoites compared to unimmunized mice. Among immunized mice 95% (19 of 20) became infected versus 92% (22 of 24) of unimmunized mice. Uninfected and infected immunized mice developed high antibody titers to the epitope after 5 booster injections and about 80-90 days. The authors found no significant relationship between antibody titer and the result of the challenge. Southern blot analysis of the parasite DNA indicated that parasites from immunized mice contained only one of two polymorphic forms of the repeat, but duplex melting studies showed that the nucleotide sequence of the repeat domain was indistinguishable in the parasites that infected unimmunized and immunized mice. The authors conclude that parasites infecting either group of mice contained the same epitopes.
