In vivo testing of subunit vaccines against malaria sporozoites using a rodent system.

| description: |
The peptide is derived from repeats N-terminal to the central repeat domain of the circumsporozoite protein.
Mice of strains BALB/c, B6.D2, and B10.D2 immunized with the epitope were not significantly protected from infection after challenge with 500 live sporozoites compared to unimmunized mice. Among immunized mice 83% (5 of 6) became infected versus 92% (22 of 24) of unimmunized mice. Uninfected and infected immunized mice developed high antibody titers to the epitope after 5 booster injections and about 80-90 days. The authors found no significant relationship between antibody titer and the result of the challenge. Southern blot and duplex melting analyses of the parasite DNA indicated that the nucleotide sequence of the repeat domain was indistinguishable in the parasites that infected unimmunized and immunized mice. The authors conclude that parasites infecting either group of mice contained the same epitopes.
