The residue numbers Lys35 and Lys130 corresponds to the positions in the mature protein.
Lys35 was determined to be a critical residue in the epitope for MAb MA-7H11. Mutations at Lys35 were selected from a randomized library of Sak-phage variants by negative selection of binding to immobilized MAb MA-7H11 and positive selection on immobilized plasmin. Analyses by surface plasmon resonance showed that soluble Sak mutants with single changes of Lys35 to Glu or Ala reduced binding to MAbs MA-7H11, MA-25E1, and MA-40C8 without significantly affecting binding to other nonoverlapping anti-Sak MAbs. The Sak mutants bound to and activated plasminogen.
