Tyr62 was determined to contribute moderately to the epitope of MAb MA-3G10 Mutations at Tyr62 were selected from a randomized library of Sak-phage variants by negative selection of binding to immobilized MAb MA-3G10 and positive selection on immobilized plasmin. Analyses by surface plasmon resonance showed that soluble Sak mutants with single changes of Tyr62 to Cys or Ala reduced binding to MAbs defining one epitope cluster (MA-3G10, MA-17G11, MA-26A2, MA-30A2, and MA-2B12) <10-fold, without significantly affecting binding to other nonoverlapping anti-Sak MAbs. Binding to plasminogen was not affected, but activation of plasminogen was eliminated.
