PEPSCAN analysis was performed to evaluate the binding of mAbs to octapeptides spanning the length of the MSP-1 protein. This peptide was not bound by any of the four mAbs. Site-directed mutagenesis of a GST-MSP-119 construct revealed critical residues for the binding of each mAbs: 12.8 = E26, R20, E24, K29, L31, K40; mAb 7.5 = N15, Q14, R20, K29, Y34, Q6, E43; mAb 12.10 = Y84 and mAb 1E1 = K40, L31.
