This peptide mimics epitopes on Plasmodium falciparum ring-infected erythrocyte surface antigen (RESA). The motif (S/T)AVDD seems to be important for binding to the monoclonal antibody 18/2 since it also occurs in other mimotopes (that bind to 18/2).
The monoclonal antibody, 18/2, raised against the C-terminal (residues 893-1073) of ring infected erythrocyte surface antigen (RESA) from Plasmodium falciparum, captured the mimotope from a random peptide phage display library. Subsequently, the mimotope was shown to bind to 18/2 by ELISA and western blot as linear or cyclized peptides, as well as after reduction and alkylation. The mimotope inhibited the binding of 18/2 to a recombinant form of RESA (aa 322-1073) as well as to infected erythrocytes, and is itself inhibited by the RESA repeats (EENV)8, (EENVEHDA)4 and (DDEHVEEPTVA)2.
