The circumsporozoite protein contains multiple tandem repeats of the sequence NANP.
The epitope was recognized by sera from rabbits immunized with a construct containing the epitope within the sequence (NANP)26 derived from the P. falciparum circumspororzoite protein and containing the C-terminal 293 amino acids of the major merozoite surface antigen (PMMSA). Immunization using Freund's adjuvant produced the highest titer antisera. The response to the epitope after the first immunization with baculovirus-expressed antigen was not increased after boosting, whereas a boosting response to a bacteria-expressed antigen was observed. For the ELISA capture antigen, the CSP peptide was synthesized with an N-terminal Cys residue and coupled to ovalbumin using glutaraldehyde.
