IgG and IgM responses to the epitope were studied after infection with E coli expressing the epitope. An IgM response was detected when the epitope was inserted within the surface expressed LamB protein, but not when it was inserted within the soluble periplasm expressed MalE protein. An IgG response was detected with the use of either LamB or MalE proteins, however, MalE required several injections to achieve an antibody response. Depletion of CD4+ T cells had little effect. Administration of the epitope within the E coli proteins rather than through the use of live E coli resulted in predominantly an IgG response which was CD4 dependent. The position of insertion within the E coli proteins played a greater role in the adminisitraion of live E coli as compared to the administration of purified proteins.
