The circumsporozoite protein contains multiple repeats of the sequence NANP.
The epitope was shown to be the minimum epitope recognized by mAb 5G5.3 by analysis of binding to a set of 4 to 8 amino acid peptides spanning the dominant repeat of the P. falciparum circumsporozoite protein. MAb 5G5.3 recognizes all octapeptides from the central repeat region, containing the dominant NANP or variant NVDP repeat. The authors deduce that 5G5.3 also binds, with less affinity, to the sequence PNVDP contained in octapeptides from the central repeat region. MAb 5G5.3 also binds to proline-containing octapeptides spaninng residues 298-311 (HNMPNDPNRNVDEN) and 361-371 (IKPGSANKPKD), outside the central repeat region.
