The terminal disaccharide of LOS-III is defined as the epitopic region for anti-LOS-III Abs. Its name is 3,6-dideoxy-4-C-(1,3-dimethoxy-4,5,6,7-tetrahydroxy-heptyl)-a-xylo-hexp-(1-3)-b-L-Xylp. The Lipooligosaccharide III (LOS III) is the source of the epitope. It is the major LOS species found in M.gastri. Its complete oligosaccharide part is 3,6-dideoxy-4-C-(1,3-dimethoxy-4,5,6,7-tetrahydroxy-heptyl)-a-xylo-hexp-(1-3)-[b-L-Xylp-(1-4)]6-3-O-Me-Rhap-(1-3)-b-D-Galp-(1-3)-b-D-Glcp-(1-4)-a-D-Glcp-(1-1)-a-D-Glcp. The lipid chains are linked to the 2 proximal 1-1 linked Glc residues.
Anti-M.gastri antibodies are inhibited in the binding to solid-pahse LOS-III by the soluble LOS-III. Based on the reactivity to other LOS molecules, the involvement of the epitopic residues is deduced.
