The epitope was capable of inhibiting the binding of preS2(ayw)-beta-gal with rabbit HBsAg-specific antisera. The determination of inhibitor concentrations, at which the binding of preS2(ayw)-beta-gal was reduced to 50%, allowed the calculation of the average antibody affinity constant (K) [3.2x107 1/mol]. A further analysis of affinity constants using a double-antibody assay confirmed the subtype specificity of the antisera-epitope intreaction.
