The epitope was selected in a screen of a 12-mer phage display library using antibodies from M. paratuberculosis-infected cattle. The antibodies were affinity-purified with a recombinant C-terminal fragment (p-34cx) of the M. paratuberculosis p34 membrane protein, in which residues 290-295 (SSPSGP) share a similar motif with the epitope (TTxxGP).
