The peptide was made cyclic by formation of a S-S bridge between the 2 Cys residues. Also, a Lys residue was added to the N-terminus for linking purposes.
MIce were immunized with this peptide (or with a longer version, with 6 extra residues at the N-ter) in different adjuvants, with similar results in the recognition of HBsAg by the AUSAB radioimmunoassay in all groups, including the controls immunized with denatured HBsAg. These fingding suggest that a single dose without carrier of a structurally constrained peptide is able to elicit a significant response, comparable to that of the complete protein, with no major impact of the adjuvant used.
