This peptide is conserved among the D/ayw2 and D/ayw3 genosubtypes. A/adw2 genosubtype differs in a single substitution, F6L, in the corresponding sequence.
By a combination of peptide ELISA, fragment WB, and trypsin cleavage of Middle HBsAg, the epitope recognized by mAb 2-12F2 was mapped to these residues. Similar reactivity towards the A/adw2 HBsAg was seen, suggesting the F6L variation does not affect the epitope recognition.
