The authors deduce that the complete epitope of mAb 3105 is discontinuous, involving residues 77-80 and 83-84 on the immunodominant loop of both subunits that form a spike on the T=4 HBV capsid. Binding of one Fab 3105 at the tip of a spike occludes the other epitope. Cyro-electron microscopy density maps of Fab 3105 bound to reassembled capsids of expressed HBV core domain were placed into high-resolution models of the capsid protein dimer onto which atomic models of Fab molecules were docked to identify the epitope. MAb 3105 was negative in Western blots of HBV capsid protein as predicted for a conformational epitope.
