The mAb 18/7 generated against the larger preS1 21-47 (PLGFFPDHQLDPAFGANSNNPDWDFNPGK) bound preferentially to the epitope present in the wild-type HBV preS1. MAb 18/7 failed to bind mutant preS1 having the altered sequence ALQPDF. Moreover, the mutant having the scrambled QLDPAF to ALQPDF motif showed poor attachment to hepatocytes cells following conjugation to beads in BMA assays. This suggests that the epitope is the site that dominates HBV attachment and mAb binding. This motif is also present in the HBV X protein.
