A yeast-display system was used to identify residues involved in binding to the conformation-dependent mAb S25 by loss of binding to mutants, where BoNT/A1 Hc residues were replaced by the respective BoNT/A2 residues based on the fact that this mAb fails to recognize BoNT/A2.
