These residues are located in the C-terminal subdomain of the binding (Hc) domain of BoNT/A, adjacent to the ganglioside binding site. This is the BoNT/A1 toxin.
A yeast-display system was used to identify residues involved in binding to the conformation-dependent mAb S25 by loss of binding to mutants, where BoNT/A1 Hc residues were replaced by the respective BoNT/A2 residues based on the fact that this mAb fails to recognize BoNT/A2.
