Binding of the IgG from haemophilia A patients to factor VIII was inhibited by the epitope as measured by Bethesda assay. Residues R6, C11 and E12 were found to be important for epitope inhibition by glycine substitution. The patient IgG also bound the epitope in direct binding ELISA.
