Testoterone-specific 77 Fab cross-reacted weakly with 5alpha-dihydrotestosterone (5alpha-DHT), which inhibited antibody binding to immobilized testosterone-3-carboxymethyloxime-BSA by 7.9%. Selection of mutant 77 Fab increased the affinity for testosterone and decreased the cross-reaction with the 5alpha-DHT about 2-fold and with DHEAS almost 3-fold in comparison with parental mutant clone A60/HCDR1/HCDR2.
