The authors deduced that the N-terminus of the TSHR ectodomain was important for mAb binding because the mAb did not bind to a chimeric TSH-LH receptor construct with residues 25-30 substituted with rat LHR residues 32-37 (PEPCD). Substitution of TSHR residues 261-418 with the LHR sequence did not significantly reduce binding. At lower Ab concentrations, the mAb preferentially bound to the GPI-anchored TSHR ectodomain expressed in CHO cells compared to the expressed wild type TSHR.
