The epitope of ABT007 Fab on the human EPO receptor (EPOR) was determined from the crystal structure of the complex. EPOR exists as a preformed dimer, however, monomeric soluble domain of the receptor was used for crystallization. The crystallographic unit [PDB: 2JIX] contains 3 EPOR monomers (chains B, C, E), and 3 Fabs: (1) chains A (light) and F (heavy), (2) G (light) and D (heavy), and (3) L (light) and H (heavy). The Fabs interact with EPOR as follows: Fab(1) with C chain (both Fab chains contact) and B (light chain contacts only); Fab(2) with chain E (both Fab chains contact) and chain C (light chain contacts only); Fab(3) with chain E (light chain contacts only). Since the contacts are different, two complexes were curated: Fab(1) - chain C and Fab(2) - chain E.
