The epitope-specific anti-A2 monoclonal antibody mAb413 demonstrated a dose-dependent inhibition of thrombin-catalyzed activation of FVIII. In a separate assay, time-course of heavy chain cleavage analyzed by blotting showed that with mAb413, cleavage at R372 was markedly diminished. This was consistent with the effect of mAb413 on thrombin-catalyzed activation of the pro-co-factor and indicate that the region comprising the anti-A2 mAb epitope is likely to contain with the thrombin-interactive site(s) that affects the cleavage at R372. in the heavy chain. The Ab also inhibited the binding of A2 to Ah-thrombin.
