The synthetic peptide C6 representing a mimotope of an epitope on the FVIII A2 domain and isolated with plasma from a hemophilia A patient blocked binding of anti-FVIII antibodies from patients’ plasma to immobilized FVIII in a concentration-dependent manner. A control peptide C6A, which is homologous to C6 except for four N-terminal amino acids, decreased IgG binding to immobilized FVIII by 40%. The addition of heterologous mimotopes increased inhibition.
