A panel of purified IgG samples derived from APS patients was selected. Direct binding (10/10) and inhibition of binding (8/8) was observed. The epitope was deduced using mutants of the wild-type domain I sequence. The greatest reduction in binding compared with wild-type domain I was observed with the single-point mutant R39S.
