The epitope-specific mAb bound to recombinant MP, but did not bind to TMV movement protein in the cell wall and ER fractions isolated from N. benthamiana cells infected with TMV, not did it recognize MP extracted from N. benthamiana or N. tabacum cells. Mutational analysis revealed that residues S1, T4, S8, and S10 were critical for binding. Phosphorylation of MP decreased the response.
