The epitope residues were calculated based on the 4 Å
atomic distance between the antigen and the antibody from [PDB: 1NBY]. The lysine residues at positions 696 (96) and 697 (97) (residues 114 and 115 in GenBank # 126608) were individually substituted with alanine.
The epitope of HyHEL-63 bound to hen egg white lysozyme, with alanine substituted residue K96A (A696) (HEL K96A; GI: 29726605), was solved from the crystal structure by molecular replacement. The affinity of HyHEL-63 for HEL K96A was reduced approximately 33,000-fold (KA = 1.1 x 104 1/M) compared to wild type HEL (KA = 3.6 x 108 1/M).
