The binding affinity of scFv FITC-E2 for fluorescein was determined by fluorescence titration. Alanine scanning of the heavy and light chain CDR regions determined the residue substitutions that resulted in a loss of binding, only some of which corresponded to contact residues in the scFV-hapten crystal structure.
