The peptide is a mimic of globotriaosylceramide from Shiga toxin.
A Gb3-trisaccharide mimic peptide was selected with biopanning from a phage display library against anti-Gb3 antibody. The binding of the selected phage clones to anti-Gb3 antibody was assayed by ELISA either with the phage clones alone, or with the clones in the presence of an excess amount of Gb3 to confirm Gb3 mimicry. In further studies, the peptide was found to bind strongly to Shiga toxin, though the binding was inhibited with Gb3. Neutralization of Shiga toxin in the presence of the peptide was confirmed by in vitro assay with HeLa cells.
