The epitope sequence was deduced through study of overlapping peptides spanning the sequence of Ara h 3. Single amino acid alanine substitutions established Glu residues at positions 6, 9 and 10, Tyr at position 7, Asp at position 8, and Arg at position 12 to be critical for IgE binding. The core epitope was determined to be YEYDEEDRR.
