The epitope sequence was determined through study of overlapping 15mer peptides spanning the antigenic region, residues 57-111. The epitope was considered immunodominant since it was recognized by sera from 6 of 7 soybean-sensitive individuals. Single amino acid alanine substitutions at positions 7, 10, and 13-15 abloished IgE binding to the peptide. Structural modeling of G2 glycinin based on the known 3-D structure of beta-phaseolin predict the epitope to be located on the surface of the trimeric G2 molecule.
