The epitope sequence was determined through study of overlapping 15mer peptides spanning the antigenic region, residues 449-471. The epitope was considered immunodominant since it was recognized by sera from 5 of 7 soybean-sensitive individuals. Single amino acid alanine substitutions did not affect IgE binding. Structural modeling of G2 glycinin based on the known 3-D structure of beta-phaseolin predict the epitope to be located on the surface of the trimeric G2 molecule.
