Monoclonal antibody raised against 12S particles containing VP1, VP2 and VP3 of FMDV type O neutralized the O epitope as presented by recombinant FMDV strain A12. The FMDV O1 BFS strain epitope described was substituted for aa 857-881 (TNKYSASGSGVRGDFGSLAPRVARQ) in the VP1 region of the polyprotein of the A12 virus. Monoclonal antibody 12FE9 was previously shown in the reference Stave et al. (1986) J Gen Virol 67:2083 [PMID: 2428926] to bind to the synthetic peptide SRNAVPNVRGDLQVLAQKVARTLPTSFNYGAIKATR corresponding to aa 861-896 in the VP1 region of FMDV strain O1 Campos. The region of overlap for mAb interaction would likely limit the epitope to aa 861-882 of the O1 VP1 polyprotein. . Note that there is one amino acid difference between the O1 BFS sequence and the original O1 Campos sequence, an exchange of L for V at position 12 of the epitope.
