description:
Since mAb R310 shares sequence homology with anti-DNA antibodies, the reactivity towards DNA was investigated. It was found that the antibody bound calf thymus ds-DNA but not synthetic oligonucletides, and therefore a natural modification of DNA was suspected as the cross-reacting epitope. By analyzing different aldehyde analogs of the eliciting immunogen as modification of DNA, it was found that recognition was extremely enhanced with 4-oxo-2-nonenal (ONE) as the modification. Different deoxy-monoribonucleotides (G,C, and A) were tested as adducts with ONE, and all were highly reactive with the mAb in ELISA, while the non-modified nucleotides were not recognized.
