The epitope is a construct of sequences from FMDV that form loop structures and comprise an antigenic site, including VP1 (193-195, TGD), VP3 (K84), VP3 (58-60, ENV), VP3 (Y63), VP2 (72-80,PSQNFGHMHK), and a VP2 analog (133-137, SEKDR).
One of 4 guinea pigs immunized with the cyclic epitope was protected from from challenge with FMDV and developed only a single minute lesion on a paw. One of 3 guinea pigs immunized with the cyclic epitope with an insertion of a 6-aminohexanoic acid residue instead of Gly next to the C-terminal Cys was completely protected and a second animal had only minor lesions. Guinea pigs immunized with the cyclic epitope modified by insertion of a 6-aminohexanoic acid or beta-Ala residue next to the N-terminal Cys, were not protected.
