| name: |
Rational dissection of binding surfaces for mimicking of discontinuous antigenic sites.
|
| description: |
The epitope is a construct of sequences from FMDV that form loop structures and comprise an antigenic site, including VP1 (193-195, TGD), VP3 (K84), VP3 (58-60, ENV), VP3 (Y63), VP2 (72-80,PSQNFGHMHK), and a VP2 analog (133-137, SEKDR).
MAbs 2A12, 2E5,3E9, and 5C4, which recognize conformational epitopes, reacted with the linear epitope or the cyclic peptide, oxidized to form an intrachain disulfide bond. Insertion of a 6-aminohexanoic acid residue instead of Gly next to the C-terminal Cys improved antibody recognition, especially of the cyclic peptide.
