description:
MAbs 23FB4 (IgG3), 23MF4 (IgG2b), and 23OG2 (IgG3) neutralized the virus in a plaque reduction assay and in a mouse protection assay, and bound to purified A5 SP-86 virions by RIA. The critical epitopic residue recognized by the mAbs was determined by sequence analysis of neutralization-resistant variants selected with the mAbs. Variants with mutation D72N in VP2 were not neutralized or bound by the mAbs. Variants with mutation E79G or E79V in VP2, selected with a different mAb, were not neutralized by the mAbs, but were still bound by the antibodies. The authors conclude D72 and E79 are part of related epitopes in the same antigenic site.
