IgM fractions from 6 infants with symptomatic congenital syphilis were tested with overlapping 8-mers to define the regions of interest. Further definition of this epitope with shorter peptides was subsequently done and this 7-mer was the optimal peptide. Alanine replacement of each residue showed that residues Phe at position 4 and Glu at position 6 were essential for antibody binding, while the alanine replacement of K2 residue enhanced binding.
