The epitopic region was mapped using two Vaccinia virus WR C-terminal deletion mutants. The epitope-specific monoclonal antibody was also shown to cross-react to several orthopoxviruses, including several vaccinia virus strains (Dryvax, Lister, IHDJ), rabbitpox virus, cowpox virus, and ectromelia virus. The epitopic region in cowpox virus (Brighton) contains a single amino acid difference when compared to vaccinia virus strain WR at residue 181 (lysine to threonine). The authors suggest the low reactivity may mean that this amino acid is important for antibody binding.
