The epitope of F8.12.19 on the P. vivax AMA1 protein was determined from the crystal structure of the complex. Only the 34-residue segment (Ile421 to Lys454) of the complete PvAMA1 ectoplasmic region could be identified in the electron density. Superposition of the complete structure of the PvAMA1 ectoplasmic region [PDB:1W8K] onto the region seen in the electron density shows that the intact antigen cannot be accommodated in the crystal lattice of the structure, suggesting that parts of the antigen were removed by proteolysis prior to crystal formation. No additional residues of the cognate antigen could be within contact distance from F8.12.19, suggesting that this epitope is a complete epitope recognized by the Fab on PvAMA1.
