MAB c78.5A recognizes a conformation-dependent epitope in domain 1 (residues 1-88) of human ICAM-1 as shown by binding to human-mouse chimeric molecules consisting of human domain 1 and the remainder of mouse ICAM-1. The critical residues for antibody recognition were shown to be P70, D71, and G72. Binding of mAb c78.5A to ICAM-1 was reduced by the single mutation D71G and the double mutation replacing residues 71 and 72 (DG) with TV, and abolished by the triple mutation replacing residues 70-72 (PDG) with GTV. These substitutions also reduced binding of human rhinovirus 3 to cells expressing the mutant ICAM-1.
