name:
Mimotope mapping as a complementary strategy to define allergen IgE-epitopes: peach Pru p 3 allergen as a model.
description:
Sequences such as this peptide were obtained by panning a synthetic library with anti-Pru p 3 allergen antisera. They conform to the consensus sequence PSRTPRPEWAXL, which was mapped onto the Pru p 3 molecule, by selecting residues located at the protein surface that exhibit proper electrostatic and solvent exposure features and choosing analogous amino acids to replace those absent in Pru p 3, to the residues L37, R39, T40, P42, D43, R44, A46, P70, S76, P78 and Y79.
A purified Pru p 3-specific IgE fraction was obtaiened by affinity selection from a pool of sera. It was used to screen a random display peptide phage library to select IgE-binding phage clones. The reactivity to this selected clone was confirmed in a phage ELISA.
