The epitope sequence recognized by serum IgE from shrimp-allergic subjects was determined in a study of overlapping peptides. The epitope core sequence ESKIVELEEEL was recognized by one serum, whereas the other 2 sera recognized the longer epitope or ESKIVELEEELRV. Critical positions for antibody recognition were determined by analysis of the epitope substituted with amino acids found in the homologous sequence from nonallergenic vertebrate tropomyosins. Amino acid substitution V191S or V199N at positions 5 and 13, of the epitope sequence, respectively, abolished IgE reactivity of 2 sera.
