Fab 57P reacted with the epitope in a spot blot of overlapping 15-mer peptides spanning residues 122-158 of TMVP. The epitope core shared by positive peptides was residues 139-145 (YNRSSFE). Amino acid substitutions indicated that residues N140, R141, and F144 at positions 4, 5, and 8 of the epitope are critical for antibody binding and showed reduced binding with all substitutions. Residue S143 at position 7 was previously shown to be essential for binding.
