This peptide is located in the variable region 2 (VR2) at the apex of the loop 4, and is part of the P1.16 epitopic determinant. The meningococcal reference strain H44/76 with B:15:P1.7,16 phenotype has the epitopes for the subtype P1.7 and P1.16 specific monoclonal antibodies in the VR1 and VR2, respectively.
The reference strain H44/76 and an isogenic strain derived from it that harbors a single residue deletion outside the epitope were equally well recognized by the epitope-specific antibody. Similar recognition is seen to a strain that differ at the epitope (D at position 2 mutated to N), whereas replacement by H abolished reactivity.
