The epitope sequence recognized by serum IgE from shrimp-allergic subjects was determined in a study of overlapping peptides. The epitope core sequence was DEERM of the peptides (RSLSDEERM, SLSDEERM, LSDEERMDALEN, SDEERMDA, DEERMDALE) recognized by sera from 6 of 10 patients. Critical positions for antibody recognition were determined by analysis of the epitope substituted with amino acids found in the homologous sequence from nonallergenic vertebrate tropomyosins. Amino acid substitution R140K at positions 4 of the epitope sequence abolished IgE reactivity of the 2 of the 6 sera.
