The peptide is conserved in other isolates, such as HCV-H, HCV-JH, HCV-J and HCV-BK, from genotypes 1a and 1b of HCV.
To determine the residues important for binding, inhibition ELISAs were performed where the epitope-specific responses were competed with peptides with terminal deletions. It was seen that the octamer PEGRTWAQ was essential for binding. By residue substitutions, A77 and Q78 were shown to be dispensable for the response in three donors, though, as substitutions to G and K (the residues found in the genotype II variant of this peptide), respectively, did not abolish binding.
