The optimal epitope recognized by the mAbs was deduced through a Pepscan analysis of overlapping peptides spanning the sequence of BRSV-G. Pepscan analysis of overlapping 8- to 16-residue peptides showed the minimal epitope recognized was CEGNLACLS for mAbs 48, 49, 50, and 66, and CEGNLACL for mAb 62, and EGNLACLSL for mAb 53. Single amino acid substitutions at every position with each of 19 other amino acids indicated that residues Cys, Cys, and Leu at positions 3, 9, and 7 of the epitope were key residues. Other residues in decreasing order of importance for binding were Glu, Asn, Ala, Leu, Ser, Gly, and Leu, at positions 4, 6, 8, 10, 11, 5, and 12.
