The epitope was designed to mimic discontinuous antigenic site D of FMDV isolate C-S8c1, which encompasses residues from VP1 (Thr193), VP2 (Ser72, Asn74, His79 in the sequence PSQNFGHMHKVVLP), and VP3 (Glu58 in the sequence FLMFENVPY). The epitope D8A, which represents the VP2 and VP3 peptides linked by a polyproline spacer, was disulfide bonded to peptide D8B (CPPPPTG), which mimics the VP1 peptide.
The binding of epitope-reactive anti-D8 sera to FMDV was inhibited by FMDV site D-specific mAbs 5C4 and 2A12, demonstrating the specificity of the anti-D8 sera.
