The sequence is conserved in other H3N2 strains such as Kiev/301/94 and Panama/2007/99. The cysteine at position 5 has a carboxyamidomethyl group as a naturally ocurring post-translational modification. This fragment and the region 158-166 (GSVNSFFSR) are recognized by the same mAb and reside in close proximity atop the HA1 subunit suggesting that they form part of a discontinuous epitope.
Both the tryptic peptide where Cys at position 5 supports the naturally ocurring post-translational modification and a second form in which this modification is replaced with a beta-hydroxyethyl group (as a result of the pretreatment of the antigen with beta-hydroxyethanol ahead of digestion) are detected. The mAb reacted with two tryptic fragments of the Shangdong and Kiev strains (which have the same sequences) as detected in Maldi-TOF spectra analysis. These fragments (109-125 and 158-166) reside in close proximity atop the HA1 subunit suggesting that they form part of a discontinuous epitope.
