TCR-like Fab antibodies specific for the HLA-A2/epitope complex were selected from a large human Fab phage library using a single chain construct of HLA-A2 linked to beta2m complexed with the epitope. Fabs T3F2 and T3E3 detected the epitope complexed with HLA-A2 on the surface of APCs pulsed with the epitope, such as murine RMA-S transfected with HLA-A2, HLA-A2+ EBV-transformed B cell line JY, or human HLA-A2+ dendritic cells. Fab tetramers showed 2-log increased staining intensity.
